Production and Characterization of fluorescent ATP Sensors with Enhanced pH Stability

    Abstract:
    In this project, two ATP sensors were developed and characterized for use in synthetic vesicles: bAT-1.03, a genetically encoded FRET sensor, and ATPBP, a hybrid sensor based on the ε subunit of ATP synthase from Bacillus sp. PS3. Both were purified, and ATPBP was labeled with fluorophores. Their fluorescent response to ATP, stability under variable pH conditions, and specificity were evaluated. The results show specific and stable detection of ATP, although the labeling of ATPBP was partial. These sensors provide new tools for synthetic biology.
