Description: | La fermentació alcohòlica és un procés microbià complex que es caracteritza per una successió de diferents espècies de llevats. Llevats amb una baixa activitat fermentativa, com són Candida spp., Hanseniaspora spp., Pichia spp., Rhodotorula spp. i Kluyveromyces spp., són predominants en el most i durant els primers dies de fermentació. Posteriorment, Saccharomyces cerevisiae prolifera, domina i completa la fermentació alcohòlica. Els llevats anomenats no-Saccharomyces spp. poden contribuir a les propietats aromàtiques i composició química del vi resultant. No obstant, aquests llevats poden ser tant beneficiosos com perjudicials. Per a un bon control del procés de vinificació, els cellers necessiten mètodes ràpids, senzills i fiables per a la identificació i tipificació de llevats a l'igual que per detectar i enumerar llevats.Els objectius d'aquesta tesi eren: (i) desenvolupar tècniques moleculars per a la caracterització, identificació i quantificació llevats vínics, (ii) avaluar la utilitat d'aquestes tècniques per identificar i quantificar llevats aïllats de mostres reals de fermentacions alcohòliques. Amb aquesta finalitat, vam utilitzar els oligonucleòtids basats en repeticions d'elements palindròmics extragèniques (REP) i repeticions d'elements consensus entobacterials intergènics (ERIC) descrits en bacteris per caracteritzar soques de llevats de referència. També vam avaluar la utilitat dels primers complementaris a ISS (intron splice site) amb el mateix objectiu. També vam desenvolupar la PCR a temps real o PCR quantitativa (QPCR) per detectar i quantificar de manera ràpida el nombre total de llevats, Saccharomyces spp. i Hanseniaspora spp. en vi sense cultiu en placa i també vam realitzar la reacció de la transcriptasa inversa i posteriorment QPCR (RT-QPCR) a p Alcoholic fermentation is a complex microbial process that is characterized by a succession of different species of yeasts. Yeasts with low fermentation activity, such as Candida spp., Hanseniaspora spp., Pichia spp., Rhodotorula spp. and Kluyveromyces spp., are predominant in grape musts and during the early stages of fermentation. Subsequently, Saccharomyces cerevisiae proliferates, dominates and completes the wine fermentation. Non-Saccharomyces spp. can contribute to the aroma properties and chemical composition of the resulting wine. However, they can be both beneficial and harmful. For a good control of the wine making process, cellars need fast, simple and reliable methods for identifying and typing yeasts as well as for detecting and enumerating yeasts.The objectives of this thesis were: (i) to develop molecular techniques for characterization, identification and quantification of wine yeasts, (ii) to evaluate the usefulness of these techniques to identify and quantify yeasts isolated from natural samples of wine fermentation. For this purpose, we used the oligonucleotides based on the repetitive extragenic palindromic (REP) and enterobacterial repetitive intergenic consensus (ERIC) elements described in bacteria to characterize reference yeast strains. We also evaluated the usefulness of the complementary primers to ISS (intron splice site) for the same objective. We also developed real-time, or quantitative, PCR (QPCR) to rapidly detect and quantify the total number of yeasts, Saccharomyces spp. and Hanseniaspora spp. in wine without culturing and we also performed a reverse transcriptase QPCR (RT-QPCR) assay from rRNA for total viable yeast quantification.
