Abstract:
Science advances constantly and bioengineering with it. As scientists we have a duty to look for methods that improve the system around us. Currently the development of new therapeutic strategies is based on animal models and in vitro two-dimensional cell cultures. But the use of animals always involves ethical issues and two-dimensional structures are not able to reproduce the physiological microenvironment of the human body. In this work, the three-dimensional culture of human myoblasts has been optimized, with the aim of synthesizing differentiated muscle tissue from patients myoblasts, with a physiological structure comparable to that of human muscle. All this has been done in order to create a new technique to reduce and if possible eliminate, the use of animals during the preclinical phase of new drugs for myotonic dystrophy type 1. This is a minority neuromuscular disease, genetic autosomal dominant, multisystemic where the most affected organ is muscle tissue, which to this day still has no cure. There are many variables to consider in order to achieve a threedimensional culture, capable of reproducing the physiology of a real muscle. We have optimized the material of the structure, the shape of the structure, the concentration of cells, the differentiation medium and the viability of the culture. All these optimizations have been verified by molecular, morphological and functional analyzes.
