Abstract:
The pathogenesis of nonalcoholic fatty liver disease (NAFLD) is triggered through multiple causes (high-fat diet, sedentary life, insulin resistance), some of them unknown. Succinate has been observed to trigger hepatic fibrogenesis through SUCNR1 signaling, but its effects on other NAFLD-associated phenomena – fibrosis and inflammation – is not yet fully described. A study from IISPV found succinate to activate an anti-inflammatory program in macrophages. In this sense, succinate-SUCNR1 could be involved in hepatic inflammation in NAFLD. This hypothesis was addressed through evaluation of inflammation, ER stress, autophagy and metabolism alterations in SUCNR1 knock-out NAFLD mice, concluding with evidence that SUCNR1 deficiency does not affect NAFLD development.
