Development of electrochemical biosensors and solid-phase amplification methods for the detection of human papillomavirus genes

A rapid, accurate and reliable diagnosis is crucial for the identification of a disease, like cancer, where an early detection can improve patient survival outcomes. Cervical cancer is the third most commonly diagnosed and the fourth leading cause of cancer death in women. It is well known that persistent infections with high-risk human papillomaviruses (HPV) are the primary cause of cervical cancer. Electrochemical DNA biosensors have received important attention owing to their characteristics. Moreover, their compatibility with microfabrication technologies makes them attractive for DNA diagnostics. The first objective of the work described in this thesis is the development of an electrochemical DNA sensor array for the multiplex detection of high-risk HPV sequences (HPV16, 18 and 45) and its application to human clinical samples. A second objective is the demonstration of a proof-of-concept of the development of isothermal (helicase-dependent amplification) solid-phase amplification methods with electrochemical real-time monitoring. Un diagnòstic ràpid, precís i fiable és crucial per a la identificació d'una malaltia, com el càncer, on la detecció precoç pot millorar els resultants del tractament. El càncer de coll uterí és el tercer més comunament diagnosticat i la quarta causa de mort per càncer en les dones. La infecció persistent de virus del papil•loma humà d'alt risc (VPH) és la principal causa de càncer de coll uterí. Els biosensors electroquímics han rebut una important atenció a causa de les seves característiques. La seva compatibilitat amb tecnologies de microfabricació els fa atractius per al diagnòstic d'ADN. Els objectius del present treball són el desenvolupament d'un sensor electroquímic per a la detecció múltiple de seqüències del VPH d'alt risc (VPH16, 18 i 45) i la aplicació en mostres clíniques. Un segon objectiu és el desenvolupament de mètodes d'amplificació d’ADN en fase sòlida amb monitorització en temps real mitjançant tècniques electroquímiques.
