Abstract:The aim of this Bachelor thesis is to optimize the technique of determination and quantification of non-starch polysaccharides used in the laboratory of IRTA-Animal Nutrition and at the same time to implement it in a new gas chromatograph. To achieve these goals, several tests have been carried out in different feeds. On the one hand, certain modifications found in different bibliography have been applied to reduce time in the preparation of the samples and on the other hand, different tests have been performed to obtain the method used in the gas chromatograph, with the corresponding calibration table to quantify samples. In addition, an attempt to introduce the use of internal standard has been made. Once the results were obtained, it was observed that the repeatability of the results in the different tests showed good. The fourth test was chosen as the most appropriate because the area percentage of the determined monosaccharides was more assimilated to that of the original methodology. Regarding to the calibration of the gas chromatograph, the third methodology was chosen because better results were obtained. The internal standard could not be implemented as the concentration mentioned in articles was too dilute. The results obtained show that the optimized method provides similar results than the previous with a short time. However, it will be necessary to continue studying other types of samples such as intestinal contents, excreta, etc. in order to see if the results obtained are the same.
