    This doctoral thesis has been done at the Unitat d'Histologia i Neurobiologia of the Faculty of Medicine at the Rovira i Virgili University. It is the result of 4 years working in the synaptic plasticity field, specifically in the loss of nerve connections.The loss of nerve connections is a process that occurs during the ageing of the nervous system and in certain neurodegenerative diseases. The process also takes place during postnatal development in a physiological way when a precise pattern of neural connections is established thanks to the elimination of redundant synapses. With this study we try to know which is the system or systems implied in the loss of nerve connections that occurs during the postnatal development at the neuromuscular junction (NMJ) -the synapses between a motoneuron and a skeletal muscle fiber-. Individual skeletal muscle fibers in newborn rodents are innervated at a single endplate by several motor axons, whereas in adult mammals, each muscle fiber is innervated by a single motoneuron. This drastic reduction in polyneuronal innervation is produced by an activity-dependent competitive withdrawal of supernumerary axons. The proteases have been implicated in this process. In this thesis I tested the hypothesis whether the maturation of the NMJ during the synapse elimination process results from a signal transduction cascade initiated by activation of the protease thrombin receptor PAR-1, localized in the postsynaptic membrane (skeletal muscle fiber) and a Protein Kinase C (PKC) activity, specifically by the theta PKC isoform activity. We have demonstrated that thrombin, through the activation of its receptor PAR-1 localized, at least in part, on the postsynaptic membrane and a theta PKC activity mediates the maturation of the neuromuscular junct
