Impact of triglyceride-rich lipoproteins on the inflammatory function of monocytes

    Abstract:
    Recent works highlight the effect of triglycerides-rich lipoproteins (TRL) in the pathogenesis of atherosclerosis. Our objective is to analyse the TRL’s effects isolated from 72 metabolic patients on the inflammatory status of macrophages. Pools of four growing quartiles of TG-TRL were incubated with THP-1 macrophages at different times to study mRNA expression and protein secretion of IL-1β (pro-inflammatory) and CD206 (anti-inflammatory). TG-TRL quartiles induced an early M1 polarization of macrophages, suggesting a pro-inflammatory effect. Reduction of IL-1β expression and later CD206 secretion propose a transcriptional regulatory effect via M2 polarization. Results support TRL’s role on the inflammatory response of macrophages.
