    Abstract:
    Protein-protein interactions between gephyrin and glycine receptors (GlyRs) allow GlyRs clustering at the postsynaptic membrane. Here, we explore the interactions of GlyR α2 subunit with gephyrin, expecting an autonomous and specific binding. Protein-peptide interactions can be evaluated at amino acid resolution by binding and neutralisation assays on peptide microarrays. As main results, the use of GlyR α2-derived peptides for competition points towards a longer sequence, gephyrin binding hierarchy shows GlyR α2 as the strongest binder among α subunits, and profiling of gephyrin-GlyR α2 interaction revealed a partially overlapping binding site. Single point gephyrin mutants revealed specifically GlyR α2 binding.
