The role of PNPLA3 protein expression in obesity-associated nonalcoholic fatty liver disease (NAFLD)

Resum: NAFLD is the most prevalent chronic liver disease in the human history (25%). It can progress to nonalcoholic steatohepatitis (NASH) characterized by fibrosis; and cirrhosis, which requires a liver transplantation. It’s indispensable a liver biopsy for the diagnosis of NAFLD/NASH progression, and there isn’t any accepted drug therapy. One of the proteins related to NAFLD is PNPLA3 (patatin-like phospholipase domain-containing 3), an enzyme involved in the lipid metabolism. PNPLA3 accumulation results in a larger lipid droplet size, characteristic of NASH. There is a need to find new targets to stop NAFLD progress, and PNPLA3 could be one of them.; NAFLD és la malaltia hepàtica crònica més freqüent en la història de la humanitat (25%). Pot progressar a esteatohepatitis no alcohòlica (NASH) caracteritzada per fibrosi; i la cirrosi, que requereix un trasplantament de fetge. És indispensable una biòpsia hepàtica per al diagnòstic de la progressió de NAFLD/NASH, i no hi ha cap tractament farmacològic acceptat. Una de les proteïnes relacionades amb NAFLD és PNPLA3 (patatina-like phospholipase domain-containing 3), un enzim implicat en el metabolisme dels lípids. L'acumulació de PNPLA3 dóna lloc a una mida de gota de lípids més gran, característica de la NASH. Cal trobar nous objectius per aturar el progrés del NAFLD, i PNPLA3 podria ser un d'ells.
