Unveiling the role of oxidized myelin debris in BV-2 microglia: Insights into multiple sclerosis

    Abstract:
    Multiple sclerosis is an autoimmune inflammatory neurodegenerative disease characterized by an immune attack on the myelin sheath, leading to an inflammatory environment with increased reactive oxygen species and myelin debris generation that must be cleared by microglia cells. Phagocytosis of oxidized myelin debris by microglia was examined using BV-2 cells. The impact on cell viability, protein synthesis, and inflammatory cytokine expression was assessed. Phagocytic activity was analyzed through CFSE staining of the myelin. Results reveal disparities in antioxidant and cellular metabolism proteins. Significant differences in phagosomal degradation were finded suggesting that oxidized myelin disrupts microglial functions and activity.
