Experimental evaluation and validation of PCR as a diagnostic tool complementary to culture for the detection of Legionella spp. in environmental water samples

    Abstract:
    Legionella spp. is a waterborne pathogen that can cause severe respiratory infections. This study evaluated whether PCR could serve as a complementary or alternative method to traditional culture for detecting Legionella spp. in water samples. Thirty samples from various sources were analyzed using both techniques. Diagnostic parameters, including sensitivity, specificity, and predictive values, were calculated. PCR showed 100% sensitivity and negative predictive value, and 63% specificity. Statistical analysis confirmed a significant association (p = 0.0006) and moderate agreement (Kappa = 0.564). PCR proved especially useful in contaminated samples, supporting its use in environmental surveillance programs.
