Desarrollo de métodos de biología molecular para el análisis directo de bacterias acéticas de vinagre

    Acetic acid bacteria (AAB) are Gram-negative, ellipsoidal that have an obligatory aerobic metabolism. They can use such substrates as glucose y ethanol as energy sources.In general, there are not many antecedents of the behavior acetic acid bacteria in natural or industrial environments. The Acetic acid Bacteria (AAB) are considered fastidious microorganisms due to their lack of good cultivability in suitable solid media. The correct application of the culture-independent methods depends mainly on a good extraction of the DNA of vinegar and wine matrices from a qualitative and quantitative point of view for their culturing. Fast and reliable methods were used to this research for identify AAB as RFLPs-PCR 16S rRNA, RFLPs-PCR ITS 16S-23S rRNA and the culture-independent methods as DGGE-PCR, general gPCR and specific gPCR with TaqMan-MGB probes. These methods were a great help to improve our knowledge of AAB.
