Determination of succinate in culture media by fluorometry

    Abstract:
    Succinate is a metabolite of the Krebs cycle that is involved in the synthesis of ATP and can act as a sensor of the metabolic state. High levels of succinate have been observed in obesity and diabetes. Obesity has been associated with intestinal dysbiosis. The microbiota has been proposed as a possible source of blood succinate. The aims of this work are to define O.laneus, C.leptum and P.xylaniphila bacteria as producers or consumers of succinate and establish a methodology in order to determine the culture media by fluorometry. A.acidipropionici, P.copri and C.butyricum, examples of consumer or producer of succinate were used as controls. The methodology performed was efficient in determining the concentration of succinate in the culture media and O.laneus was characterized as a succinate producer. C. leptum and P.xylaniphila were not characterized.
