Expression and purification of the leptin receptor extracellular domain

Abstract:
The leptin receptor remains a challenge in the structural biology field leading to the lack of high-resolution structure data. This research is aimed to perform the expression and purification of different constructs of its extracellular domain to work towards reaching an expression protocol that gives enough protein levels for crystallography. A eukaryotic cell expression system that allowed the expression of different domains of the extracellular region of the leptin receptor linked to purification tags that allow to isolate them was used. Low-yield expression of some of the recombinant proteins was achieved, however, the purification protocol must be further improved.
