Experimental Approaches for Detecting Long Noncoding RNAs in Malignant Pleural Effusion

    Abstract:
    This study focuses on establishing the optimal experimental conditions for detecting long noncoding RNAs (lncRNAs) which are tumour-derived products present in pleural fluid (PF). A qPCR assay was conducted to evaluate the impact of different initial sample volumes, the inclusion of an RNA spike-in (cel-miR-39) during the extraction process and the implementation of a preamplification step. The results suggest that the optimal conditions for lncRNA extraction from PF involve an initial volume of 200μL, the addition of cel-miR-39 and carrying out of a preamplification procedure. Furthermore, ACTB and GAPDH were validated as reference genes for data normalisation in PF samples.
