Development and optimization of a minimal-volume 1h-nmr protocol for lipoprotein quantification in clinical and research contexts

    Abstract:
    This study aimed to develop and validate a reduced-volume ¹H-NMR protocol for lipoprotein quantification without compromising analytical quality. Lipoproteins, key in lipid transport and cardiovascular risk assessment, are typically measured using the Liposcale test, which requires larger sample volumes. The proposed protocol was tested on nine serum samples classified by lipid concentration. Statistical methods, including the Wilcoxon test, Bland-Altman plots, and Z-scores, were used to assess precision and reproducibility. Results suggest the modified protocol is feasible, though with some limitations. The study concludes that lipoprotein profiling can be optimized for minimal-volume samples, but broader validation is needed in larger cohorts.
