Hybrid wheat production: characterization of a cytoplasmic male sterility and fertility restoration system based on Hordeum chilense chromosome 6 introgression into the Triticum durum genome

    Abstract:
    This thesis investigates the potential of the T6HchS·6AL translocation from Hordeum chilense to restore fertility in cytoplasmic male-sterile (CMS) durum wheat. Molecular markers and genomic in situ hybridization confirmed stable integration of the alien chromatin into the wheat genome. Chromosome spreads were optimized using nitrous oxide treatment, and cytogenetic stability of the CMS line was verified. Although controlled crosses were attempted, fertility restoration could not be assessed due to mildew infection and poor flowering synchrony. Nonetheless, the validated T6HchS·6AL line is a step toward hybrid wheat breeding using H. chilense introgressions.
