Analythical method development by HPLC-DAD for squalene determination in DOD samples

    Abstract:
    Clariant has just started a project in order to create a new plant, which will produce squalene. Squalene is a compound which demand is increasing in recent years in the cosmetics industry. Because it is considered to have interesting properties as anti-aging effect, hydration, and so on. The aim of this project is to develop a method to determinate the squalene’s content in the raw material samples by liquid chromatography (HPLC-DAD). The raw material from which squalene is obtained is DOD, a byproduct obtained during the stages of refining olive oil process. We took a reference method as a starting point to begin to make the first tests. Some parameters were modified in order to find the right chromatographic conditions. Once the optimal conditions were found, it was created a calibration curve with the purpose to quantify the future sample that will be analyzed. The results were compared with another method (GC-FID) which was developed at the same time that this to know if the results were comparable. Finally, some statistical parameters were studied to validate the method.
