Developement and validation of bioactive compounds analysis method in vegetables by HPLC

    Abstract:
    Animal nutrition industry has been investigating different bioactive compounds which have antioxidant, antiviral and antimicrobial characteristics in order to reduce the use of antibiotics in nutrition mixtures, and then avoid antimicrobial resistances and also avoid multiresistant bacteria generation. Vegetables are rich sources of bioactive compounds, in particular, phenolic acids, flavones, flavanones, flavanols and, in some cases, limonoids. In this work, a method was developed and validated for determination of these polyphenolic compounds from four vegetable matrices. All samples were treated with ethanol using ASE (accelerated solvent extraction) as an extractor to extract these bioactive compounds. Analyses were performed by liquid chromatography, HPLC, using DAD (diode array detector) and MS (TOF, time of flight) as a detector. This method was validated according to the requirements for new methods, which include selectivity, linearity, precision and accuracy, limit of detection and limit of quantitation.
