Identification of new interaction partners for Aquaporin 4 in S. cerevisiae using BiFC and FACS

    Abstract:
    Aquaporin 4 (AQP4) is strongly important for water transport in the human brain. In healthy conditions, it is polarized in perivascular astrocyte end foot membranes. However, interactions with certain proteins may affect that polarization, leading to neuropathological conditions. In this project, new interaction partners of AQP4 are searched for in a human brain cDNA library using Saccharomyces cerevisiae as host by coupling Bimolecular Fluorescence Complementation and Fluorescence-Activated Cell Sorting. Positive results have been obtained that may relate to an unknown protein interacting with AQP4 in the brain. Further analysis must be carried out to elucidate the nature of that interaction.
